研究簡介:
神經元的型態以及它們所伸展出來的神經纖維 -- 軸突與樹突 -- 的形狀,可以告訴我們它所擔任的功能嗎?我們可由認識神經元在腦部區域的位置、型態,將神經元加以分類並且了解它們可能扮演的不同功能與角色。利用果蠅為實驗對象,我們針對與學習和記憶有關的基因和神經元在大腦網絡的相對位置與關係來做研究。如果所有表現與學習和記憶有關基因的神經元,在腦中的位置一一被辨識並且繪製成網絡圖譜,同時了解基因產物之間的交互作用,將可以有助於我們透視大腦運作的機制。
利用我們發展的專利影像技術,現在可以很清晰地獲取並建構出表達與學習和記憶有關基因的神經元位置,和由其所組合而成的整個果蠅大腦神經網絡圖譜,並且經由結合多項技術如遺傳工程、轉殖綠色螢光蛋白的突變株果蠅製備、活體影像擷取、免疫染色組織技術、鈣離子顯像、多光子共軛焦點顯微鏡、電子顯微鏡以及三維立體影像分析的研究成果,將此一功能性圖譜稱之為記憶網絡圖譜。我們的長期目標是建立一個可以存取果蠅記憶圖譜的數位化資訊的資料庫,同時含有細胞與分子層次方面的資訊,以供研究者對果蠅大腦的運作有詳盡的了解。
